In a move considered one of the most significant recent developments in cancer treatment, Merck and Moderna announced promising preliminary results from a personalized experimental vaccine based on messenger RNA (mRNA) technology, as part of Phase III clinical trials—the critical stage preceding formal regulatory approval.

The vaccine targets patients with high-risk cutaneous melanoma who have undergone complete tumor resection. The therapy was tested on more than 1,100 patients within a treatment regimen combining the personalized vaccine with Merck’s Keytruda immunotherapy.

Strong Results in Extending Recurrence-Free Survival

Data showed that combining the vaccine with Keytruda significantly prolonged patients’ recurrence-free survival compared to using Keytruda alone, while also reducing the likelihood of cancer spreading to other organs in the body. This represents a pivotal advance in treating melanoma, the most lethal form of skin cancer despite its relative rarity.

These findings reinforce earlier results from Phase II trials, offering hope that this therapeutic approach could become a new option for patients living with the anxiety of recurrence during the first few years after treatment.

A Personalized Approach Targeting Each Patient’s Mutations

Dr. Jane Hiley, Head of Oncology Development at Merck, explained that the vaccine is designed individually for each patient, targeting the specific genetic mutations of the tumor rather than following a one-size-fits-all treatment approach. The vaccine trains the immune system to recognize and eliminate these mutations, while Keytruda enhances the body’s ability to attack cancer cells.

Hiley noted that side effects were similar to those associated with traditional vaccines, emphasizing that the results hold significant implications not only for melanoma patients but for cancer patients in general. The vaccine is also being tested in other trials involving non-small cell lung cancer, bladder cancer, and renal cell carcinoma.

A Step Toward Treating Multiple Cancers

Analysts believe these results may pave the way for the vaccine’s approval to treat various types of cancer, opening the door to a new era of personalized therapies tailored to each patient’s genetic profile.

The two companies plan to present the data at an upcoming international medical conference, while the timeline for submitting regulatory approval applications in the United States has not yet been determined.
