Caffeinated Coffee or Decaffeinated: Which Is Better for Brain Protection?

A recent study conducted on laboratory mice revealed that consuming caffeine-containing coffee protects the brain from damage caused by chronic stress, whereas decaffeinated coffee showed no similar protective effect.
The journal *Neurobiology of Stress* published the results of this study, which suggested that caffeine specifically, rather than other coffee components, is primarily responsible for its mood- and memory-protective properties.
Researchers Angelo Tomé and Rodrigo Cunha from the University of Coimbra in Portugal led a team aiming to resolve a long-standing debate regarding caffeine’s specific role in resisting stress effects. They did so by directly comparing the impacts of regular coffee versus decaffeinated coffee on mice exposed to unpredictable chronic stress, a laboratory model used to simulate symptoms of depression and anxiety in humans.
The researchers divided 24 adult mice into four groups: one group drank water without being subjected to any stress, while three other groups underwent three weeks of variable stress. During this period, the stressed groups drank either water alone, an extract of caffeine-containing coffee, or an extract of decaffeinated coffee. According to the study, the amount of coffee consumed voluntarily by the mice was equivalent to an adult human consuming approximately 350 milligrams of caffeine daily, which corresponds to two to three cups of coffee.
The results showed that mice that drank only water under stress lost weight, exhibited excessive anxiety, displayed clear deficits in spatial memory, and lost interest in pleasurable activities—symptoms scientifically known as anhedonia. In contrast, mice that consumed caffeinated coffee maintained behavior nearly identical to the non-stressed control group; they did not lose weight and showed no signs of anxiety or memory impairment.
Histological analysis of the brain revealed that stress caused a sharp decline in long-term potentiation within the hippocampus and reduced levels of brain-derived neurotrophic factor in the prefrontal cortex in the mice that drank water. Conversely, caffeinated coffee provided complete protection for these two neural mechanisms, while decaffeinated coffee failed to offer any significant protection.
These findings align with previous research covered by *PsyPost* in 2024 and 2025, which demonstrated that caffeine protects rodents from stress-induced spatial memory disorders and maintains chemical balance in the brain. However, the researchers highlighted two main limitations in the current study:
• The exact levels of caffeine circulating in the mice’s blood and tissues were not measured, as the animals were housed in groups to avoid isolation stress.
• The tests were conducted on a mixed group of males and females without isolating the gender factor, despite previous studies suggesting potential gender differences in coffee’s effects on mood.
The researchers emphasized the need for caution when generalizing these results to humans, given the significant differences in dietary habits, stressors, and brain chemistry between humans and rodents. They affirmed that the study opens the door to broader clinical research on the protective role of caffeine against disorders associated with chronic stress.
