Long Sleep Signals Alzheimer's

A new study conducted by researchers at the University of Texas Health Science Center at San Antonio found that consistently sleeping for too many hours is associated with elevated levels of a protein linked to Alzheimer’s disease in the blood, even after accounting for other influential health factors. The study relied on data from more than 2,410 participants in the Framingham Heart Study.

The researchers observed a relationship between sleep duration and levels of phosphorylated tau181 protein, a modified form of tau protein that serves as a distinctive biological marker for Alzheimer’s disease and can now be directly detected in blood samples. Using advanced non-linear mathematical models, the team discovered that levels of this protein begin to rise noticeably when usual sleep duration reaches between 8.5 and 9 hours per night, with the increase accelerating sharply after surpassing the 10-hour threshold.

Vanessa Young, a research fellow at the Glenn Biggs Institute for Alzheimer’s and Neurodegenerative Diseases, noted that many people are concerned about how their sleep habits affect brain health. She emphasized that the use of non-linear models was essential to uncovering this relationship, stating that “forcing a straight line onto data that is inherently curved causes us to completely miss the essence of the story.”

• Specificity of tau181 protein: The team simultaneously tracked four different proteins associated with cellular damage and neurodegeneration. However, the association between long sleep duration and three of these proteins disappeared once the results were adjusted for kidney function. Only the tau181 protein remained significant after this statistical adjustment, reinforcing its status as a specific indicator closely tied to Alzheimer’s disease pathways.

The researchers concluded that long sleep duration may not be the direct cause of this protein’s accumulation, but rather a behavioral indicator reflecting silent neurodegenerative processes occurring at a very early stage. This opens the door to using sleep duration as a supportive criterion for the early detection of Alzheimer’s disease in future studies.
